چکیده

Rationale: The ability to perform on the Cambridge Neuropsychological Test Automated Battery touchscreen pairedassociate learning (PAL) test is predictive of Alzheimer’s disease and Mild Cognitive Impairment. Recently, an automated computer touchscreen PAL task for mice has been developed. Pharmacological validation of this task is warranted to establish it as a useful tool in future drug discovery pertaining to Alzheimer’s disease and Mild Cognitive Impairment. Objectives: This investigation provides a systematic analysis of nicotinic involvement within the PAL task for mice. Particularly, the effects of systemic administration of nicotinic cholinergic agents (agonist and antagonist) on PAL task performance in C57BL/6 mice were investigated. This was done to detect whether bidirectional modification of performance is consequent upon these manipulations. Methods: Upon acquiring the PAL task, nicotine (nicotinic receptor agonist; 0.1, 0.5, and 1.0 mg/kg) and mecamylamine (nicotinic receptor antagonist; 0.3, 1.0, and 3.0 mg/kg) were administered intraperitoneally to the mice in a withinsubjects design, prior to daily sessions in the PAL task. Results: Nicotine did not have any significant effect on PAL performance improvement at any doses. However, mecamylamine did increase perseverative responding and reaction time in the mice. Such impairment effects are interpreted as being attentional in nature. Conclusion: This investigation indicates that mice indeed acquire the rodent PAL task, deeming it a valuable tool for future drug discovery. Further, the nicotinic cholinergic system appears to be implicated in PAL task performance, with greater effects seen with deactivation rather than activation of the system, and with these effects appearing to be of an attentional nature.

Glucocorticoids Interact with Cholinergic System in Impairing Memory Reconsolidation of an Inhibitory Avoidance Task in Mice

Introduction: Recent studies suggest that glucocorticoids modulate memory reconsolidation. Moreover, cholinergic system is involved in memory reconsolidation. Since glucocorticoids interact with brain cholinergic system in modulating memory processing, we investigated whether glucocorticoid influences on the reconsolidation of emotionally arousing training depend on the cholinergic system...

Effect of the cholinergic and opioid receptor mechanisms on nicotine-induced analgesia

In this study, we investigated the effect of nicotinic receptor agonists and antagonists on the analgesic response to morphine in the formalin test. In experiments conducted in mice, nicotine produced an early dose-dependent analgesic effect. At a dose of 0.5 mg/kg, mecamylamine, a nicotinic receptor inhibitor, suppressed the analgesic effect induced by 0.1 mg/kg nicotine in both stages of th...

Varenicline Ameliorates Learning and Memory Deficits in Amyloid β(25–35) Rat Model of Alzheimer’s Disease

Introduction: Alzheimer’s disease (AD) is a enfeeble neurodegenerative disorder characterized by increased β-amyloid (Aβ) deposition and neuronal dysfunction leading to impaired learning and recall. Among proposed risk factors, impaired cholinergic transmission is a main cause for incidence of disease. Methods: In the present study, effects of the intracerebroventricularly administration of an ...

The role of the dorsal hippocampus in two versions of the touchscreen automated paired associates learning (PAL) task for mice

Rationale The CANTAB object-location paired-associate learning (PAL) test can detect cognitive deficits in schizophrenia and Alzheimer’s disease. A rodent version of touch screen PAL (dPAL) has been developed, but the underlying neural mechanisms are not fully understood. Although there is evidence that inactivation of the hippocampus following training leads to impairments in rats, this has no...

Role of the thalamic parafascicular nucleus cholinergic system in the modulation of acute corneal nociception in rats

The present study investigated the effects of microinjections of acetylcholine (a cholinergic agonist), physostigmine (a cholinesterase inhibitor), atropine (an antagonist of muscarinic cholinergic receptors) and hexamethonium (an antagonist of nicotinic cholinergic receptors) into the parafascicular nucleus of thalamus on the acute corneal nociception in rats. Acute corneal nociception was ind...
